Potensi Pertumbuhan Beberapa Bakteri Simbion Larva Oryctes Rhinoceros pada Media Nutrient Agar dan Pengaruh Derajat Keasaman (pH) Terhadap Pertumbuhan Bakteri pada Media Nutrient Broth
Potential Growth of Several Symbiont Bacteria of Oryctes Rhinoceros Larvae on Nutrient Agar Media and the Influence of Acidity Level (pH) on Bacterial Growth on Nutrient Broth
Abstract
One type of pest that adversely affects the growth of oil palm trees is Oryctes rhinoceros. O. rhinoceros larvae are among the insects that harbor symbiotic bacteria. This study aims to characterize the biochemical and morphological aspects of symbiotic bacteria from the digestive tract of O. rhinoceros larvae. on oil palm stems affected by Ganoderma fungus infection, to determine the growth potential of symbiotic bacteria from the digestive tract of O. rhinoceros larvae on Nutrient Agar media in vitro, and to assess the effect of pH on the growth of O. rhinoceros larval symbiotic bacteria on Nutrient Broth media. This research was conducted at the Plant Disease Laboratory, Faculty of Agriculture, Universitas Sumatera Utara from November to May 2024. This research was conducted using a Completely Randomized Non-Factorial Design method with 3 replications to study the effect of pH levels on bacterial growth in Nutrient Broth and bacterial colony growth on Nutrient Agar. The best bacterial growth on Nutrient Agar was observed in bacteria A, with the average total population at 48 hours being 7.16 x 10⁸. The best treatment was observed in treatment A3 (Bacteria A in Nutrient Broth with pH 7) and B3 (Bacteria B in Nutrient Broth with pH 7).
