Isolation and Identification of Flavonoid Compound from Nam-Nam Stem Bark (Cynometra Caulifora L.)

Isolation of flavonoid compound from stem bark of nam-nam (Cynometra
caulifora L.) had been carried out through the extracted maceration with methanol
solvent, then re-extracted with ethyl acetate, partitioned extracted with n-hexane and
obtained methanol concentrated extract. The concentrated methanol extract was then
analyzed by TLC, then separated by column chromatography with silica gel as the
stationary phase and chloroform:ethyl acetate (90:10, 80:20, 70: 30, 60:40, 50:50,
40:60, 30: 70, 20:80, 10:90) v
/v as mobile phase. The pure compound obtained was
in the form of a yellow amorphous solid, mass=10 mg with Rf=0,55 using the eluent
chloroform: ethyl acetate (20:80) v
/v. Based on UV-Vis spectrophotometer analysis
with acetone solvent, it shows that there are two absorption peaks with wavelengths
(λmax) of 350 nm and 255 nm. The FT-IR spectrum shows the presence of O-H, C=O
ketone, C=C aromatic, C-O-C asymmetric and symetric bending group and C-H sp2
groups respectively at the wave number 3410,15; 1658,78; 1606,70; 1261,45;
1122,57; 873,11 cm-1
. Proton Nuclear Magnetic Resonance Spectrum (1H-NMR)
shows the presence of H-3, H-6, H-8, H- 2', H-6' protons. Spectroscopic data shows
that the isolated compound is flavone group
