The aim of the study was to determine the effect of concentration and duration of colchicine immersion on the growth of peanut embryos (Arachis hypogaea L.) in vitro. This research was conducted at the Alifa Agricultural Research Center Laboratory (ALIFA-ARC) and the Physiology and Plant Tissue Culture Laboratory, Faculty of Mathematics and Natural Sciences, University of North Sumatera, from October 2023 to January 2024. This Completely Randomized Design (RAL) was used with two factors. The first factor was the concentration of colchicine (0 ppm, 50 ppm and 100 ppm). The second factor was colchicine soaking time (0 hours, 12 hours and 24 hours). The parameters observed were the percentage of live explants, planlet length, number of leaves, number of buds, wet weight, stem diameter, stomatal size and stomatal density. The results showed that the concentration of colchicine significantly affected the parameters of the number of leaves, number of buds, wet weight and stem diameter. Duration of colchicine soaking significantly affected the number of leaves, number of buds, wet weight and stem diameter. The interaction between colchicine concentration and soaking time not significant affected Based on the results showed that the colchicine with a concentration of 50 ppm with a soaking time of 12 hours could increase the growth of peanut embryos in vitro.
