The Effectiveness Test of Tiger Gagatan Leaf Nanoherbs (Vitis gracilis Wall.) Against Testicular Histology of Wistar Rats (Rattus norvegicus L.) Male Model of Alloxan-Induced Diabetes Mellitus
Abstract
Diabetes mellitus can affect the testicular organs, including a reduction in the size and weight of the testicular size and weight, increased abnormalities in spermatogenesis, characterised by the number of sperm produced. Gagatan harimau leaves contain secondary metabolites, namely flavonoids as antioxidants, which can improve the histology of the testicular organs. The aim of this study was to determine the efficacy of gagatan harimau leaf nanoherbs on the morphology and histology of the testes of male Wistar rats (Rattus norvegicus L.), which are models of alloxan-induced diabetes. This study used a completely randomized design (CRD) with 6 treatments and 4 replicates. Negative control was normal rat treatment. Positive control of diabetic rats injected with alloxan dose (120 mg/kg bw), treatment of diabetic rats with standard drug treatment of glibenclamide (0.45 mg/kg bw), treatment of diabetic rats with treatment of gagatan harimau leaf dose 100, 125, 150 mg/kg BB. After 14 days of treatment, the rats were necropsied and the testicular organs were harvested and prepared by the paraffin method with hematoxylin and eosin (HE) staining. The results showed that gagatan harimau leaf nanoherb (Vitis gracilis Wall.) at a dose of 125 mg/kg bw can improve morphology and reduce damage (p <0.05) including seminiferous tubule diameter, germinal epithelium thickness of seminiferous tubules, Leydig cells, spermatogonium, spermatocytes and spermatids.
