Research on fish diversity was conducted in the waters of Poncan Gadang Island, Central Tapanuli Regency, North Sumatra in March 2023. The purpose of this study was to analyze fish diversity and its relationship with physical-chemical factors in the waters of Poncan Gadang Island. The research was conducted using a purposive sampling method at 3 different stations based on the activities around the island waters. Fish samples were taken using nets at each station and then identified. The physical chemical parameters measured included temperature, pH, salinity, light penetration, light intensity, oxygen saturation, dissolved oxygen, BOD5, NO3 and PO4. The analysis included fish density, relative density, frequency of presence, diversity index, uniformity index and correlation index of macroobenthic diversity to physical and chemical factors in the waters. The research results obtained were 2 orders, 14 families, and 21 genera. Fish with the highest density was found at station 1 with a value of 0.13 ind/m2 by Stolephorus indicus. The highest diversity index was at station 2 of 2.752 and the highest uniformity index was at station 2 with an index of 0.904. Correlation values between the fish diversity index and the physico-chemical factors of the waters which had a very strong effect are salinity, DO, BOD and oxygen saturation on the fish diversity index in the waters of Poncan Gadang Island.
