Isolation and Evaluate Potential of Laccase-Producing Endophytic Fungi from Oil Palm (Elaeis guineensis Jacq.) Roots
Abstract
Endophytic fungi from oil palm (Elaeis guineensis Jacq.) known produce degradative enzymes such as laccase. This study aimed to obtain endophytic fungi produce laccase enzymes isolated from oil palm roots continue by identifing potential isolate. The isolates obtained were characterized macroscopically and microscopically under the light microscope, and selected their ability to produce laccase enzymes based on qualitative methods. Quantitative methods also under taken by measuring activity of enzyme and spesific activity of enzyme produce by fungi on PDB medium and PDB + corn cob powder medium. The potential isolate with the highest laccase activity identified molecularly using the primers ITS1 and ITS4. There are 5 isolates potential in produce laccase marked by red color merged on the colony JES 01, JES 04, JES 05, JES 06, and JES 09. JES 01 shown the highest laccase activity. Laccase enzyme produced on PDB medium show the higher specific activity of enzyme compare with laccase enzyme produce growth on PDB + corn cob powder medium. JES 01 95,12% similar with the Marasmius palmivorus voucher SFSU:DED 8219 type.
