    Abstract
    Background: The development of spectrophotometric methods for determining the mixtures of two or more drug components with chromophore groups has advanced rapidly. This rapid advancement is due to the progress of computer technology combined with practical mathematical calculations applied in software. The determination of vitamin C and quercetin levels using the Mean Centering Ratio (MCR) method is one of the spectrophotometric methods can be used to determine simultaneously drug levels in combination without separation. Objective: To confirm the Mean Centering Ratio (MCR) method can be used and meets the method validation requirements for the simultaneous determination of vitamin C and quercetin levels in tablets using ultraviolet spectrophotometry. Method: The determination of vitamin C and quercetin levels using ultraviolet spectrophotometry with the Mean Centering Ratio (MCR) method involved several steps, including the determine absorption ratio spectra, MCR spectra, calibration, and method validation using MCR. The method validation parameters tested included linearity, accuracy, precision, limit of detection (LOD), and limit of quantitation (LOQ). Results: The results showed that Mean Centering Ratio (MCR) method meets the validation requirements. The average content of vitamin C and quercetin in Life Extension® dissolved in methanol solvent were (100.05 ± 1.16)% for vitamin C and (93.29 ± 0.92)% for quercetin. Conclusion: Based on the results of research, the levels of vitamin C and quercetin in tablets determined using the Mean Centering Ratio (MCR) method has met the drug levels requirements in Indonesian Pharmacopoeia VI edition (2020) and this method has met the method validation requirements according to International Conference on Harmonisation (ICH) guidelines.
