    Abstract
    Background: Ketoprofen is a non-steroidal anti-inflammatory drug (NSAID) that has poor solubility that can cause irritation and gastrointestinal disorders. Xanthan gum is one of the natural polymers that provides good characteristics and stability. Therefore, ketoprofen is formulated into a nanosuspension preparation with the addition of natural polymer xanthan gum. Objective: To formulate ketoprofen nanosuspension preparation with xanthan gum polymer, characterize the preparation and compare the dissolution test with ketoprofen suspension. Method: Preparation of ketoprofen nanosuspension using evaporativeprecipitation method with Tween 80 surfactant and variation of xanthan gum polymer concentration F1 (0.05%), F2 (0.25%), F3 (0.5%). The characterization of the preparation includes organoleptic test, determination of particle size, zeta potential, and polydispersity index, measurement of pH, specific gravity, viscosity, content and dissolution test of the preparation, and observing the stability of the preparation for 3 months at room temperature storage (25C). Results: Ketoprofen can be formulated into nanosuspension with xanthan gum polymer. The particle sizes of ketoprofen nanosuspension in F1, F2, F3 are 15,620,24 nm; 16,740,17 nm; 18,640,36 nm, with zeta potential >-30mV and polydispersity index value >0.2. The preparation has a pH of 4.6-5.0; specific gravity >1 g/mL; and viscosity of 20-32 cP. The preparation showed stability during 3 months storage. However, there was a decrease in pH, particle size, zeta potential, polydispersity index, specific gravity, viscosity and content. The dissolution results with % cumulative at the 60th minute were 83.75%; 81.28%; 80.18% respectively, showing faster results than ketoprofen suspension with % cumulative 12.13%. Conclusion: Ketoprofen nanosuspension can be made with gom xanthan polymer. There is an effect of concentration variation in gom xanthan polymer on the characterization of ketoprofen nanosuspension preparation with the best nanosuspension results at a concentration of 0.05%. The formulated nanosuspension showed faster release compared to ketoprofen suspension.
