    Abstract
    Background: Matoa is an endemic Indonesian plant originated from Papua. Matoa contains secondary metabolites that have various properties for the body, such as phenols and flavonoids. Phenol and flavonoids have activity as antioxidants. Efforts to maximize the inclusion of nutrients contained in matoa leaf can be done by reducing the size of matoa leaves to the size of nanoparticles. Objective: This study aims to determine of total phenols, total flavonoids, and antioxidant activity of matoa leaf nano powder. Methods: The particle size of matoa leaf powder was reduced by Planetary Ball Mill and measured by Particle Size Analyzer. Then, phenol and flavonoid screening was conducted on matoa leaf nano powder. Matoa leaf nano powder was dissolved in methanol to 1000 μg/ml, sonicated for 1 hour and left for 1 day. Determination of total phenol content was carried out by colorimetric method using Folin-Ciocalteu and Na2CO3. Determination of total flavonoid content was done by colorimetry, with AlCl3 and CH3COONa. Antioxidant activity test was conducted with DPPH method. Results: The screening results of phenol and flavonoid compounds in matoa leaf nano powder showed positive results for the presence of phenol and flavonoid compounds in matoa leaf nano powder. The total phenol content obtained was 232.3319 ± 14.66 mg GAE/g sample. Total flavonoid content obtained was 4.5288 ± 0.99 mg QE/g sample. Antioxidant activity of matoa leaf nano powder obtained IC50 value of 38.3519 ± 0.62 μg/mL. Conclusion: Matoa leaf nano powder contains phenol and flavonoid compounds. The antioxidant activity of matoa leaf nano powder falls into the very strong category.
