    Isolasi dan Penentuan Struktur Kimia Senyawa Flavonoida Daun Benalu Jeruk (Scurrula fusca G. Don) Serta Uji Aktivitas Antioksidan

    Abstract
    The aims of this research are to isolate, examine antioxidant activity and chemical structure of flavonoid compounds from citrus parasite leaves (Scurrula fusca G.Don). Flavonoid compounds isolated from the leaves of citrus parasite by extracting 3000 g leaves powder using methanol. Methanol extract re-extracted with ethyl acetate. Ethyl acetate extract was partitioned with mixture of methanol and n-hexane. Methanol layer hydrolyzed with HCl 6% and obtained 3.02 g (0.10%) aglycone of flavonoids. Aglycone of flavonoids were isolated and purified by column chromatography. Pure compounds F3 (10 mg) was obtained as a brownish green amorphous solid and F4 (14 mg) as a yellow amorphous solid. The antioxidant activity of both compounds were investigaed by DPPH free radical scavenging method using ascorbic acid as a control positive. Antioxidant activity of F3 and F4 obtained with IC50 values of 9.383 μg / ml and 8.713 gμ / ml respectively. The antioxidant activity of both compounds classified as very powerful because they have IC50 values lower than ascorbic acid antioxidant activity with IC50 value of 12.08 gμ / ml. Determination of the chemical structure of both compounds F3 and F4 done by interpretation of spectral data of spectroscophy FT-IR, UV, 1D NMR (1H NMR, 13C NMR, and DEPT),2D NMR (1H-1H1 COSY, 1H-13C HMBC, 1H-13C HMQC) dan LCMS. Based on these data, F3 expressed as 3.7-dihydroxy-5-methyl-3', 4'- dihydrofurano-flavonol and F4 as 3,5,7,4'-tetrahydroxyflavonol or Kaempferol.
     
    Tujuan penelitian ini adalah mengisolasi, menguji aktivitas antioksidan dan menentukan struktur kimia senyawa flavonoida dalam daun benalu jeruk (Scurrula fusca G. Don). Senyawa flavonoida diisolasi dari daun benalu jeruk dengan cara mengekstraksi 3000 g serbuk daun benalu jeruk menggunakan metanol. Ekstrak metanol diekstraksi kembali dengan etil asetat. Ekstrak etil asetat dipartisi dengan campuran metanol dan n-heksana. Lapisan metanol dihidrolisis dengan HCl 6% sehingga diperoleh 3,02 g (0,10%) aglikon flavonoida. Aglikon flavonoida yang diperoleh diisolasi dan dimurnikan dengan kromatografi kolom. Dua senyawa murni diperoleh, yaitu F3 yang berbentuk padatan amorf berwarna hijau kecoklatan dengan bobot 10 mg, dan F4 berbentuk padatan amorf berwarna kuning dengan bobot 14 mg. Aktivitas antioksidan kedua senyawa murni ini diuji dengan metode penangkapan radikal bebas DPPH menggunakan asam askorbat sebagai pembanding. Aktivitas antioksidan F3 dan F4 menunjukkan IC50 9,383 μg/ml dan 8,713 μg/ml berturut-turut. Aktivitas antioksidan kedua senyawa ini tergolong sangat kuat karena memiliki nilai IC50 lebih kecil dari nilai IC50 asam askorbat 12,08 μg/ml. Penentuan struktur kimia kedua isolat F3 dan F4 dilakukan dengan cara interpretasi data spektroskopi FT-IR, UV, 1D NMR (1H NMR, 13C NMR, dan DEPT), 2D NMR (1H-1H COSY, 1H-13C HMBC, 1H-13C HMQC) dan LC-MS. Berdasarkan data tersebut, isolat F3 dinyatakan sebagai 3,7-dihidroksi-5-metil-3’,4’-dihidrofurano-flavonol dan isolat F4 dinyatakan sebagai 3,5,7,4’-tetrahidroksiflavonol atau Kaempferol.
