    Studi Aktifitas Immunostimulan Daun Buas Buas (Premna pubescens. Blumue) pada Tikus Putih (Rattus norvegicus)

    Abstract
    Immunostimulant is a substance to increase the immune responsiveness. Premna pubescens.Blumue (buasbuas) has immunostimulant activity by the presence of Apigenin. Apigenin value on Premna pubescens.Blumue was 28.45mcg/ml.This research used 24 Wistar ratsdivided into 4 treatment groups which each A0 was control wich given normal food, A1 was given 250 mg Ethanol Ectract of Premna (EEP)/kg bw, A2 was given 250 mg EEP/kgbw + SRBC 1% and A3 given SRBC only.EEP was given orally for 30 days and SRBC was given on 8th and 15th day of treatment.Blood collection doneon 31th day of treatment. Imunostimulant activity done with measurement of hematology (hemoglobin, hematocrit, erythrocyte and platelet), sellular (leucocyte, lymphocyte, monocyte), humoral(antibody titer, IgM, IgG and Lisozim) and histology (kidney and spleen). For antibody titer and immunoglobulin M and G analyzed by hemaglutination and ELISA technique.Based ANOVA test, leucocyte, lymphocyte and plateletswere have a significant effect. Leucocyte and antibody titer at A2significant increase compare with A3(9.88±2.31>7.52±1.82 x103/mm3 dan 7.17±0.75>6.67±1.51). IgG and IgM have the same result that A2was the highest (3.96±and 9.48±5.90 mg/ml). For Lysozyme measurement show that A2was the highest 0.04μg/ml.Antibody titer and IgM significantly increased compared to the control. For histological overview, centrum arteriole and centrum germinal at A2 is the highest value. The whole result that EEP have immunostimulant activity, because it enhance immune responses in leucocyte, lymphocyte, platelets, antibody and IgM.
     
    Imunostimulan adalah agen kimia yang dapat meningkatkan respon imun tubuh. Premna pubescens.Blumue memiliki aktivitas sebagai imunostimulan karena adanya senyawa Apigenin. Nilai apigenin pada Premna pubescens.Blumue adalah 28.45mcg/ml. Dalam penelitian ini digunakan tikus putih jantan sebanyak 24 ekor dan dikelompokkan menjadi empat kelompok yaitu A0(Kontrol), A1 diberikan 250 mg EEP/kg bb. A2, diberi 250 mg EEP/ kg bb + 0.1 ml SRBC 1%, dan A3 hanya diberikan 0.1 ml SRBC 1%. Perlakuan diberikan setiap hari selama 30 hari dan SRBC diberikan pada hari ke 8 dan 15 perlakuan.Pengambilan sampel darah dilakukan pada hari ke-31.Aktivitas imunostimulan dilakukan dengan pengukuran terhadap hematologi, (hemoglobin, hematokrit, eritrosit dan trombosit), imunitas selular (leukosit, limfosit, monosit), imunitas humoral (titer antibody, IgM, IgG dan Lisozim) dan histologi (limpa dan ginjal). Pengukuran titer antibodi dan imunoglubulin M dan Gmenggunakan uji hemaglutinasi dan teknik ELISA.Berdasarkan uji ANOVA, leukosit, limfosit, dan trombosit meningkat signifikan. Leukosit dan titer antibodi menunjukkan nilai A2> A3 (9.88±2.31>7.52±1.82 x103/mm3 dan 7.17±0.75>6.67±1.51). Analisis kadar IgM dan IgG A2 memiliki kadar IgM dan IgG tertinggi (3.96±1.05 dan 9.48±5.90 ng/ml). Hasil pengukuran kadar lisozim diperoleh A2 memiliki kadar lisozim tertinggi 0.04μg/ml. Titer antibodi dan IgM menunjukkan adanya perbedaan yang signifikan dibandingkan dengan kontrol. Pada histologi limpa, diameter sentrum arteriol dan germinal pada A2 memiliki rata-rata nilai tertinggi. Hasil penelitian menunjukkan bahwa EEPmemiliki aktifitas imunostimulan karena dapat meningkatkan respon imun tubuh pada jumlah leukosit, limfosit, trombosit, titer antibodi dan IgM.
