    Abstract
    Background: Gadung tuber (Dioscorea hispida Dennst.) is widely spread in Indonesia, but has not been fully studied. Gadung tubers have various chemical compounds, one of which is diosgenin. Diosgenin belongs to the steroid saponin class which can be used as synthetic steroid precursors. The need for diosgenin supply in the pharmaceutical industry continues to increase so that the economic opportunity in the development of diosgenin is very profitable. Diosgenin can be obtained from isolation of gadung tuber ethanol extract. Purpose: The purpose of this study was to isolate diosgenin from gadung tuber extract and identified by UV-Visible spectrophotometry and fourier transform infrared (FTIR). Method: These study included gadung tuber simplisia production, phytochemical screening, ethanol extract, characteristics test, isolation by preparative thin layer chromatography and identification by UV-Visible spectrophotometry and fourier transform infrared. Result: The isolation from ethanol extract of gadung tuber showed a wavelength of 205 nm and 453 nm by UV-Visible spectrophotometry. Ethanol extract of gadung tuber showed several functional groups, havely -OH, C=C aromatic, and C-O aromatic when identified by FTIR. Conclusion: Based on the results of research that has been conducted, diosgenin compound can be isolated from gadung tuber ethanol extract by preparative thin layer chromatography and identification using UV-Visible spectrophotometry and fourier transform infrared showed that the isolate obtained from the ethanol extract of gadung tuber was indeed a diosgenin compound.
