    Abstract
    Background: In the determination of multicomponent levels by UV spectrophotometry, if drug compounds were obtained whose maximum wavelengths were close to each other and interfered with each other, the determination of levels was carried out by developing UV spectrophotometric methods. Determination of vitamin C and quercetin levels using chemometric techniques was one of the developments of spectrophotometric methods that could be used to determine the levels of drug compounds in mixed form. Objective: To determine the levels of vitamin C and quercetin in capsule preparations simultaneously using the UV spectrophotometric method using the chemometric technique combined with multivariate Partial Least Squares (PLS) calibration. Methods: The stages in this study include the preparation of calibration models, validation, and determination of levels. Chemometric calibration modeling was performed for each of the five concentrations of vitamin C and quercetin, and absorbance was measured at wave numbers 210–274 nm and 216–280 nm. This was followed by validation using the parameters R2, PRESS, and RMSECV, then determination of levels using the divisor factor. Results: Validation results on PLS multivariate calibration of vitamin C and quercetin with each value of RMSECV, PRESS, R2 respectively, were 0.00368 and 0.00639; 0.00043 and 0.00131; 0.99999 and 0.99999. Based on these results, it showed that the prediction ability was still acceptable and well used for level determination. The results of determining the levels of vitamin C and quercetin contained in the capsule, respectively, were 100.59 ± 0.42% and 100.13 ± 1.25%. Conclusion: Based on the results of the research that had been done, it could be concluded that the determination of vitamin C and quercetin levels using the PLS multivariate chemometric method met the level requirements of validation parameters and met the level requirements for capsule preparations according to the Indonesian Pharmacopoeia VI Edition (2020).
