    Abstract
    Potato production in Indonesia is not yet sufficient and fluctuates. One of the constraints is the limited use of land for seed propagation and the high cost of in vitro media. This study aims to obtain the optimization of the use of MS (Murashige and Skoog) medium in growing several potato varieties to form micro tubers in vitro. This study was conducted at the Biotechnology Laboratory of the Faculty of Agriculture, USU Medan at an altitude of ± 45 meters above sea level from June to November 2023. The study was conducted by planting node cuttings of four potato varieties (Granola, Dayang Sumbi, Atlantik Malang, and Maglia) on MS medium whose concentration had been optimized (½ MS, ¾ MS, and 1 MS). This study used two stages, namely the propagation stage (shoot micropropagation) and the micro tuber formation stage. The results showed that the optimization of the ¾ MS medium concentration could be used at the propagation stage seen from the number of nodes, the number of leaves, and the number of primary roots of potato plantlets. Furthermore, the optimization of the ½ MS medium concentration could be used at the potato micro tuber formation stage. In the variety treatment, Dayang Sumbi, Atlantik Malang, and Maglia gave the best response at the propagation and micro tuber formation stages. There was no interaction effect between the concentration of MS medium and several potato varieties.
