    Test of Anti-Aging Activity on Clay Mask Containing Red Spinach Leaves (Amaranthus tricolor L.) Ethanol Extract

    Abstract
    Background: Red spinach leaves (Amaranthus tricolor L.) contained antioxidant compounds such as tannins, alkaloids, saponins, flavonoids in the form of anthocyanins that function to ward off free radicals and prevent premature aging on the face. Ethanol extract of red spinach leaves (Amaranthus tricolor L.) has potential as anti-aging active ingredient of clay mask. Objective: To prove that the clay mask containing ethanol extract of red spinach leaves have anti-aging activity. Methods: This study included the stages of sample processing, simplisia characterization, extract preparation, extract characterization, phytochemical screening, and clay mask formulations with concentrations of 0.5% (F1), 1% (F2), 1.5% (F3), without ethanol extract of red spinach leaves (F0) as a blank and clay mask sold in the market (F4) as a positive control. Evaluation of clay mask preparation included stability test, homogeneity, pH, dry time of preparation test, irritation test, and anti-aging activity test. Anti-aging activity was tested during 4 weeks treatment period, with parameters of moisture, pores, blemishes, and wrinkles on the facial skin of volunteers measured once a week using a skin analyzer. Results: Phytochemical screening of simplisia and ethanol extracts of red spinach leaves showed the presence of alkaloids, flavonoids, saponins, glycosides, tannins, and steroids. The results of the evaluation of clay mask preparations were homogeneous, dry time of preparation is 10.66-18.66 minutes, pH 5.01-6.88, does not irritate the skin, and is stable in storage for 12 weeks at room temperature. The clay mask which has the best stability based on pH and organoleptic test, namely F2 (1%) showed percent recovery of moisture (31.54%), pore (48.12%), blemishes (26.25%), and wrinkles (13.54%). Conclusion: The clay mask of ethanol extract of red spinach leaves (Amaranthus tricolor L.) has good anti-aging activity based on the percent recovery of the four anti-aging parameters, namely moisture, size, blemishes, and wrinkles.
