    Abstract
Background: Acne can generally be treated by using antibiotics that can inhibit the growth or kill the bacteria. Infectious diseases are the most major health problems in developing countries such as in Indonesia Infectious diseases are caused by microorganisms such as viruses, fungi, and bacteria Propionibacterium acnes and Escherichia coli bacteria are easily resistant to antibiotics and there are many negative impacts of long-term antibiotic use, so it is necessary to develop antibacterial agents by utilizing plants, one of the plants that can be used is the leaves of the branches. Objective: The purpose of this study is to determine the antibacterial activity of ethanol extract and fraction against Propionibacterium acnes and Escherichia coli bacteria. Methods: The method used in this study is the disk diffusion method. The samples were extracted by maceration using 70% ethanol, then fractionated using ethyl acetate solvents, and n-hexane. The antibacterial activity test of the leaf fraction of rantiti uses an in vitro testing method, namely disc diffusion with concentrations of 80, 70, 60, 50 mg/ml. Results: The results of testing the antibacterial activity of ethanol extract and leaf fraction showed antibacterial activity against Propionibacterium acnes and Escherichia coli bacteria. Conclusion: Ethanol extract of rantiti leaves has antibacterial activity against the growth of Propionibacterium acnes and Escherichia coli bacteria with a moderate inhibition zone category. The n-hexane and ethyl acetate fractions of the leaves of the antiti leaf have antibacterial activity against the growth of Propionibacterium acnes and Escherichia coli bacteria with a strong inhibitory zone category. The leaf fraction that has the highest inhibition zone in bacteria is the ethyl acetate fraction with an average inhibition zone of 13.2 mm against Propionibacterium acnes bacteria and has an average inhibition zone of 12.3 mm against Escherichia coli bacteria.
